3-MB-PP1 is specific inhibitor of analogue-sensitive ZAP70 (ZAP-70(AS)), does not affect Lck or Tec kinase activity, specifically targets ZAP-70AS-dependent events.
3-MB-PP1 selectively shows sensitivity against the cells expressing the ZAP-70AS alleles.
3-MB-PP1 treatment (6 uM) blocked TCR-mediated signaling in both ZAP-70AS1 and ZAP-70AS2 stably transfected cells, with only a negligible effect on ZAP-70WT cells.
3-MB-PP1 effectively blocked calcium responses, inhibited almost all NFAT-driven luciferase activity, suppressed superantigen-mediated IL-2 induction.
