Biological Activity
BLU3797 is a highly potent, selective, orally bioavailable PIK3C2B (phosphatidylinositol-4-phosphate 3-kinase) inhibitor with IC50 of 28 nM (1 mM ATP), while sparing other PI3K family members.
BLU3797 has low affinity for other kinases in general as assessed by kinome scan, with an overall S(10) selectivity score of 0.015.
BLU3797 (3 uM) promotes myocyte-myotube fusion in C2C12 cells.
BLU3797 effectively rescues the MTM1-null phenotype
BLU3797 normalizes disease biomarkers, improves muscle function, and enhances survival in preclinical models of X-linked myotubular myopathy (XLMTM).
