BMS-684 (BMS684) is a potent, selective inhibitor of diacylglycerol kinase α(DGKα) with IC50 of 15 nM, weakly inhibist DGKβ and DGKγ, does not inhibit other DGK lipid kinases.
BMS-684 exhibits a dose-dependent potentiation of IL-2 production with EC5 of 5.43 uM.
BMS-684 does not induce a T cell response in the absence of TCR stimulation.
BMS-684 exhibits the hallmark phenotype of a candidate intracellular T cell checkpoint inhibitor.
BMS-684 induces translocation of DGKα to the membrane in T cells with EC50 of 0.54 uM in the YFP-DGKα high-content translocation assays.
