Belrestotug (EOS-448, EOS 884448, GSK 4428859, GSK 4428859A) is a fully human IgG1 TIGIT antagonist monoclonal antibody, fully prevents CD155-mediated inhibition.
EOS-448 does not cross-react with mouse TIGIT, the anti-TIGIT antibody used in murine models is a surrogate antibody produced at UProtein.
EOS-448 prevents CD155 binding and increases the functional activity of TIGIT+ cells, blocks TIGIT restored secretion of IFNγ in a dose-dependent manner on CD8+ T cells from healthy donor PBMCs activated in the presence of CD155-expressing cells, also boosts the functional activity of T cells in samples from solid tumor patients.
EOS-448 induces preferential depletion of Tregs,triggers preferential antibody-dependent lysis of Tregs ex vivo.
