Biological Activity

Butyzamide is a potent, specific non-peptidyl activator /agonist of Thrombopoietin (TPO) receptor Mpl, promotes the proliferation of Ba/F3-hMpl cells with EC50 of 10 nM.
Butyzamide does not promotes the proliferation of parental Ba/F3 cells that does not express human Mpl.
Butyzamide shows species specificity for human but not murine Mpl, Butyzamide does not stimulate other cytokine receptors of EPO, G-CSF, GM-CSF and IL-3.
Butyzamide induces phosphorylated JAK2, STAT3, STAT5 and MAPK in Ba/F3-hMpl cells.
Butyzamide induced colony-forming unit-megakaryocyte and polyploid megakaryocytes from human CD34+ hematopoietic progenitor cells.
Butyzamide (3 uM) increased polyploid megakaryocytes in a similar manner to 1 nM rhTPO.
Butyzamide (10 or 50 mg/kg once daily for 20 daysincreased human platelets in FL-CD34+-transplanted NOG mice.
