CDK2 inhibitor 5 is a potent, selective and allosteric CDK2 inhibitor with KD value of 18 nM, shows exquisite selectivity for CDK2 over CDK1.
CDK2 inhibitor 5 demonstrates negative cooperativity with cyclin A.
CDK2 inhibitor 5 is selective for CDK2 over the very similar CDK1 in this cellular context.
CDK2 inhibitor 5 does not show >80% inhibition of any kinase at a 10 µM concentration, and only 4% of the 371 kinases tested showed >50% inhibition.
CDK2 inhibitor 5 recapitulates features of Cdk2−/− and Spdya−/− phenotypes in spermatocytes.
