Biological Activity
CK-571 (CK-2018571) is a potent and selective inhibitor of Smooth muscle myosin-2 (SMM) with IC50 of 86 nM.
CK-571 specifically inhibits the steady-state actin-activated MgATPase activity of SMM with IC50 of 9 nM, over other Myosin II isoforms (cardiac and striated muscle myosins).
CK-571 also has a significantly lower affinity for nonmuscle myosin II (NMM, ∼8.5-fold less).
CK-571 binds to an allosteric pocket that becomes momentarily available during one of the intermediate steps of the recovery stroke of the myosin motor.
