Biological Activity
CLEO4-88 is a molecular glue binder of CTLH E3 ligase substrate adaptor GID4 with KD of 5.6 uM, promotes GID4 interaction with the peroxisomal thiolase protein ACAA1 with EC50 of 12.5 nM in HTRF assays, inhibits the catalytic activity of ACAA1.
CLEO4-88 makes no direct contact with ACAA1.
Ternary complex formation with GID4 and CLEO4-88 inhibits ACAA1 thiolytic activity with IC50 of 1.8 uM.
CLEO4-88 can act as a molecular glue to drive interaction of GID4 and ACAA1 in live cells.
