Biological Activity
CSN5-14 is a small molecule COP9 signalosome subunit 5 (CSN5, JAB1) inhibitor with IC50 of 0.73 uM, inhibits TNBC cell proliferation and clonogenic survival.
CSN5-14 effectively engages intracellular CSN5 and suppress its deneddylase activity in a dose- and time-dependent manner.
CSN5-14 induces dose- and time-dependent accumulation of NEDD8-Cul1 in TNBC cells, without altering the protein level of CSN5.
CSN5-14 displays potent cytotoxicity with IC50 values of 8.1 uM in MDA-MB-231 and 5.8 uM in MDA-MB-468 cells.
CSN5-14 significantly impaired clonogenic capacity in a dose-dependent manner in MDA-MB-231 cells.
CSN5-14 elevate ROS, induce cell-cycle arrest and apoptosis in TNBC cells.
