DTHIB (Direct Targeted HSF1 InhiBitor, SISU-102) is a specific small molecule HSF1 inhibitor with Kd of 160 nM for DTHIB binding to the HSF1 DNA binding domain (DBD).
HSF1 is highly expressed in human prostate cancer and is inhibited by DTHIB, suppresses AR activity and signaling.
DTHIB inhibits human prostate cancer cell proliferation (PC-3, EC50=3 uM).
DTHIB inhibits tumor growth in animal models oftherapy-resistant prostate cancer, with no weight loss or organ damage.
