DrOTOP1 inhibitor C2.2 is a potent, specific small molecule inhibitor of zebrafish (Danio rerio) Otopetrin 1 (DrOTOP1) proton-selective ion channel with IC50 of 6.67 uM in HEK-293 cells expressing DrOTOP1 (pH 5.5).
C2.2 is slightly less effective at inhibiting DrOTOP1 currents at pH 5.0, while no difference was observed between pH 6.0 and pH 5.5 (IC50 values were: pH 6.0: 5.97 µM, pH 5.5: 6.67 µM, and pH 5.0: 12.6 µM).
C2.2 fully blocked currents at 200 µM in whole-cell patch-clamp recording of DrOTOP1 expressed in HEK-293 cells.
C2.2 significantly inhibited mouse OTOP1 (mOTOP1), but not human (Homo sapiens) OTOP1 (hOTOP1).
C2.2 is specific for DrOTOP1 over mammalian orthologs.
Otopetrins (OTOPs) are a conserved, eukaryotic family of proton-selective ion channels, OTOP1 serves as a gustatory sensor for sour tastes and ammonium chloride.
