ERAP1 inhibitor N1 is a selective small molecule ERAP1 inhibitor, disrupts its function in Hedgehog (HH) signaling pathway (IC50=46.24 nM). ERAP1 inhibitor N1 inhibits the ERAP1/USP47 interaction by increasing the stability of βTrCP. ERAP1 inhibitor N1 inhibits HH-dependent cell growth of cerebellar granule neuron progenitors. ERAP1 inhibitor N1 suppresses the growth of primary HH-MB cells in vitro. ERAP1 inhibitor N1 can crosse the BBB and inhibits the growth of primary HH-MB in an orthotopic implantation model.
