Fabimycin is a potent, specific inhibitor of the enoyl-acyl carrier protein reductase enzyme FabI, exhibits impressive activity against >200 clinical isolates of Escherichia coli, Klebsiella pneumoniae (MIC, 1 ug/mL), and Acinetobacter baumannii, and does not kill commensal bacteria.
Fabimycin potently inhibits A. baumannii and E. coli FabI with IC50 of <10 nM in enzyme assays.
Fabimycin inhibited 90% of a panel of 100 K. pneumoniae clinical isolates at 4 μg/mL, relative to 32 μg/mL for levofloxacin.
Fabimycin demonstrates in vivo efficacy in murine urinary tract infection (UTI).
