Biological Activity

Glovadalen (UCB0022) is a potent, selective, brain-penetrant and orally available dopamine D1 receptor (D1R) positive allosteric modulator (PAM).
UCB0022 selectively binds D1Rs with nanomolar affinity and enhanced the potency of dopamine to activate D1Rs by approximately 10-fold.
UCB0022 is inactive at D2, D3 and D4 receptors and marginally active at D5 dopamine receptors at 10 uM.
UCB0022 improved motor disability similar to levodopa but with reduced dyskinesia in MPTP-treated macaque.
