Biological Activity
HUHS199 is a potent small molecule inhibitor of prostate cancer antigen-1 (PCA-1, ALKBH3), potently inhibits the RNA demethylase activity of ALKBH3 with IC50 of 0.17 uM in ELISA assays.
HUHS199 significantly inhibits cell proliferation in a concentration-dependent manner on U251MG GBM cells, via G1 arrest of cell cycle.
HUHS199 increases m1A levels of RNA in U251MG cells.
HUHS199 inhibits the RNA demethylase activity of ALKBH3, thereby increasing the m1A levels of GADD45A, one of its substrates, increase enhances GADD45A stability and, through interaction with p21, induces G1 phase cell cycle arrest in GBM cells.
