IB15C is a small molecule binder targeting the inhibitory immune receptor ILT3 (LILRB4, CD85k) with MST Kd of 0.92 uM, potently inhibits the ILT3-ApoE interaction with IC50 of 0.48 uM in ELISA-based competition assays.
IB15C produces a robust concentration-dependent reduction in binding signal, with an IC50 of 0.64 uM in BLI assays.
IB15C disrupts ILT3-ApoE complex formation rather than indirectly altering assay components or protein stability.
IB15C (1-10 uM) reduced SHP1 and SHP2 phosphorylation in a concentration-dependent manner.
IB15C suppresses ILT3-dependent signaling, reduces inflammatory activation, and restores Aβ uptake in human iPSC-derived microglia.
IB15C attenuates NF-κB activation, significantly reduces IL-1β secretion and restores Aβ internalization.
