Biological Activity
INCB191358 is a potent, selective, and orally bioavailable dual DGKα/ζ inhibitor with IC50 of 0.12/0.36 nM respectively, shows good selectivity over Type I DGK isoforms.
INCB191358 shows potent activity in the cellular (EC50 = 12 nM) assay and excellent activity in human whole blood (h-WB EC50 = 22 nM).
INCB191358 induced robust, dose-dependent CD8+ T-cell activation in the OT-I murine model and, in combination with anti-PD-1, produced significant antitumor activity in the MC38 syngeneic mouse model.
