A potent, selective positive allosteric modulator of mGlu2 receptor with EC50 of 17 nM; displays >100-fold selectivity over mGlu1,3-8 receptors; shows a central in vivo efficacy by inhibition of REM sleep state (3 mg/kg, po) in the rat sleep–wake EEG paradigm; reverses PCP-induced hyperlocomotion ( ED50 of 5.4 mg/kg, sc).
