K284 is a selective small molecule inhibitor of chitinase 3 like1 (CHI3L1) with strong binding affinity (Kd=-9.7 kcal·mol-1), inhibits lung metastasis by blocking IL-13Rα2-mediated JNK-AP-1 signals.
K284 inhibits the growth of A549 (IC50=2.5 uM) and H460 cells (IC50=2.7 uM), arrests cell cycle, and induces apoptotic cell death.
K284 (0.5 mg/kg) inhibits B16F10 melanoma metastasis to the lungs in C57BL/6 mice.
K284 inhibits CHI3L1/IL‐13Rα2 signaling and JNK/AP‐1 signaling in A549 and H460 cell lines.
