KKL-1005 is an anti-tubercular triazole-based molecule, binds to ribosomal protein bL12 (MST Kd=8 uM, purified E. coli bL12) and specifically inhibits the trans-translation ribosome rescue pathway, inhibits trans-translation in E. coli cells with IC50 of <1.5 uM in E. coli expressing mCherry-trpAt.

KKL-1005 inhibits M. tuberculosis trans-translation in in vitro with IC50 of 37 uM.
KKL-1005 inhibits growth of strain H37Rv by 95.5% when present at 10 µM (3.7 µg/mL).
KKL-1005 inhibits growth of the attenuated M. tuberculosis strain H37Rv ΔRD1 ΔpanCD with MIC of 4.7 ug/mL.
KKL-1005 binds the N-terminal domain of ribosomal protein bL12, binds with purified M. tuberculosis bL12 with MST Kd of 4 uM.
Overexpression of bL12 suppresses growth inhibition by KKL-1005.
