LA-CB1 is a novel Abemaciclib derivative that induces CDK4/6 degradation through the ubiquitin-proteasome pathway, exerts robust anti-proliferative effects across a wide spectrum of cancer cell lines (A498 cell, IC50=0.21 uM).
LA-CB1 inhibits triple-negative breast cancer cell line MDA-MB-468 with IC50 of 0.45 uM.
LA-CB1 (0.25, 0.5, and 1 µM) inhibits the clonogenic growth and DNA synthesis in breast cancer cells.
LA-CB1 (1 uM) inhibits migration and invasion in MDA-MB-231 and MCF-7 cells by modulating EMT-related proteins.
LA-CB1 induces cell cycle arrest and promotes apoptosis in breast cancer cells, downregulates CDK4/6 and Cyclin D1, and promotes caspase-dependent apoptosis.
LA-CB1 (0.5 mg/kg, 1 mg/kg, and 2 mg/kg) inhibits tumor growth in an orthotopic 4T1 breast cancer model.
