Biological Activity
MDI-117740 is a highly potent, selective dual-LIMK1/2 inhibitor with binding Kd of 16/76 nM respectively.
MDI-117740 displays excellent cellular target engagement and potently inhibits p-cofilin levels in SH-SY5Y cells.
MDI-117740 does not bind PAK1–2/4, ROCK1/2, MRCKα or CAMK4, RIPK1.
MDI-117740 significantly inhibits wound closure compared to untreated control at 0.25 μM and 1 μM concentrations, has a more pronounced effect observed at 3 μM.
MDI-117740 exhibits potent inhibition of cofilin phosphorylation, and positive LIMK-dependent downstream phenotypic effects on cell migration.
