ML267 (ML 267) is a potent, small-molecule inhibitor of bacterial phosphopantetheinyl transferase Sfp-PPTase with IC50 of 0.29 uM.
ML267 displays no activity toward the human orthologue (hPPtase).
ML267 attenuates production of an Sfp-PPTase-dependent metabolite when applied to Bacillus subtilis at sublethal doses.
ML267 demonstrates antibacterial activity against methicillin-resistant Staphylococcus aureus, B. subtilis (HM489, IC50=9.3 uM) and chemical genetic studies implicated efflux as a mechanism for resistance in Escherichia coli.
