	MLT-231 (MLT231) is a highly potent, selective, allosteric MALT1 inhibitor with biochemical IC50 of 9 nM, inhibits MALT1 cleavage of the BCL10 substrate with IC50 of 0.16 uM.
	MLT-231 exhibited high MALT1 selectivity over other proteases.
	MLT-231 inhibited cellular proliferation (low micromolar range) and cleavage of MALT1 substrates in ABC-DLBCL.
	MLT-231 (100 mg/kg) dose-dependently reduced the anti-DNP IgM and IgG responses induced by 2,4-dinitrophenol (DNP) in mice, blocked T cell-dependent B cell responses.
	MLT-231 (100 mg/kg, oral bid schedule for 2 weeks) exhibited antitumor efficacy in OCI-Ly10 ABC-DLBCL type xenograft models.
