Biological Activity

MNS1-MV is a novel pyrazole-based small molecular inhibitor of STAT3 pathway, potently inhibit IL-6-induced STAT3 phosphorylation at 10 μM in dBT114 cells.
MNS1-MV shows selective toxicity towards adult GBM and pediatric high grade glioma cells with IC50 values of 10.89 μM for MC-BT114 cells, 11.71 μM for MC-PED8 cells and 8.5 μM for SF7761.
MNS1-MV (25 uM) effectively inhibits STAT3 activation and nuclear translocation in both adult GBM and pediatric HGG cells.
MNS1-MV inhibits STAT3 activation via IL-6-mediated STAT3 pathway and not direct JAK2.
MNS1-MV (5-25 uM) induces apoptosis and inhibit migration in adult GBM cells.
