MSDC0160 (Mitoglitazone, MSDC-0160) is a small molecule targeting mitochondrial pyruvate carrier (MPC), also is an insulin sensitizer with anti-diabetes activities.
MSDC0160 exhibited glucose-lowering effects similar to those of pioglitazone but without the adverse effects associated with PPAR-γ agonists.
MSDC0160 has low affinity for binding and activation of PPARγ and has insulin-sensitizing effects in mouse models of diabetes.
MSDC0160 increased AMPK activity and reduced mTOR activity in human islets.
MSDC-0160 in combination with IGF-1 and 8 mM glucose increased β-cell specific gene expression of insulin, pdx1, nkx6.1, and nkx2.2, and maintained insulin content without altering glucose-stimulated insulin secretion.
MSDC0160 suppresses tumor growth in preclinical models of AR-driven PCa.
