Mtb GpsI inhibitor X1 is a specific small molecule inhibitor of mycobacterial PNPase, guanosine penta-phosphate synthase I (GpsI), with IC50 of 1.95 uM.
X1 functions as a noncompetitive inhibitor of Mtb-GpsI, with dissociation constant Ki of 0.79 uM and 0.95 uM for degradation and synthesis of poly(A).
X1 inhibits Mtb-GpsI but not Synechocystis PNPase in a dose-dependent manner.
X1 exhibits potent bactericidal properties, killing 99% of Mtb within 72 h at MIC concentration of 4.4 mg/l (7.8 μM) and within 48 h at two-fold MIC (15.6 μM).
X1 binds to the conserved phosphate-binding pocket and traps the poly(A) substrate.
