NCT-503 is a small molecule 3-phosphoglycerate dehydrogenase (PHGDH) inhibitor (IC50=2.5 uM) that reduces the production of glucose-derived serine in cells and suppress the growth of PHGDH-dependent cancer cells.
NCT-503 exhibits EC50 of 8-16 uM for the PHGDH-dependent cell lines; has reasonable aqueous solubility and favorable ADME properties.
NCT-503 selectively increases necrosis in MDA-MB-468 xenografts.
