Biological Activity
PB151 is a highly potent and isoform-selective HDAC11 inhibitor with IC50 of 59 nM, displays > 172-fold selectivity margin over all other class I, IIa, and IIb HDAC isoforms.
PB151 selectively engages HDAC11 in cells and inhibits its intracellular defatty-acylase activity.
PB151 suppresses HCC proliferation and induces apoptosis, inhibiting the growth of HepG2, MHCC97-L, and HuH-7 cells with IC50 values of 5.50 uM, 7.08 uM, and 6.67 uM respectively (72 h of exposure).
PB151 (30 mg/kg once daily, i.p.) suppresses tumor progression in an endogenous orthotopic HCC model.
PB151 treatment improves liver histology and downregulates Ki67 expression in orthotopic hepatocellular carcinoma model.
PB151 treatment alters fatty acid metabolism and reduces lipid accumulation in hepatocellular carcinoma.
PB151 restricts lipogenesis via activation of the LKB1-AMPK signaling axis.
