Biological Activity
PMM-172 is a specific small molecule STAT3 inhibitor targeting STAT3 SH2 domain, inhibits STAT3 downstream target genes expression, shows anti-proliferative activity against MDA-MB-231 cells with IC50 of 1.98 uM.
PMM-172 induced apoptosis in MDA-MB-231 cells in dose- and time- dependent manners.
PMM-172 reduced mitochondrial transmembrane potential in MDA-MB-231 cells.
PMM-172 reduced constitutive pSTAT3 (Tyr705) and inducible STAT3 activation in MDA-MB-231 cells.
PMM-172 does not inhibits phosphorylation or expression of STAT1 and STAT5.
PMM-172 suppressed STAT3 nuclear localization, and inhibited STAT3 downstream target genes expression.
