SARS-CoV-2 Mpro inhibitor 18 has 10-fold weaker inhibition of MERS Mpro compared to SARS-CoV Mpro, inhibits HCoV-229E Mpro with IC50 of 10 nM.
SARS-CoV-2 Mpro inhibitor 18 exhibits similar EC50 values across the USA-WA1/2020, alpha, beta, delta, and omicron variants (IC50<100 nM)
SARS-CoV-2 Mpro inhibitor 18 is also active against the alpha coronavirus HCoV-229E (EC50 = 0.15 μM), beta coronavirus HCoV-OC43 (EC50 = 0.15 μM), and MERS-CoV (EC50 = 0.058 μM).
SARS-CoV-2 Mpro inhibitor 18 (300 and 600 mg/kg/d, oral) significantly reduced lung virus titers, exhibited in vivo efficacy against SARS-CoV-2 MA10 infection in mice.
