SB-612111 is a potent, selective nociceptin/orphanin FQ receptor (NOP receptor) antagonist with Ki of 0.33 nM; shows weak or little affinity for μ-, κ- and δ-receptors with Ki of 57.6, 160.5 and 2109 nM, respectively; antagonizes the pronociceptive action of nociceptin in an acute pain model, and potentiates the action of morphine in morphine-tolerant animals and blocks hyperalgesia in an inflammatory pain model.
