SLM6031434 (SLM 6031434) is a potent, highly selective sphingosine kinase 2 (Sphk2) inhibitor with Ki value of 340 nM (hSphk2), 23-fold selectivity over hSphk1.
SLM6031434 inhibits mSphk2 with Ki of 400 nM, with no activity against mSphk1.
SLM6031434 dose-dependently increased Smad7 expression and ameliorated the expression of Col1, FN-1 and CTGF in primary renal fibroblasts.
SLM6031434 demonstrated anti-fibrotic potential in unilateral ureter obstruction (UUO), a model for progressive renal fibrosis.
