SM141 is a potent, selective peptidomimetic dual inhibitor of SARS-CoV-2 main protease (MPro) and host cathepsin L (CatL) with IC50 of 0.9 uM and 0.06 uM respectively.
SM141 blocks SARS-CoV2 replication in hACE2 expressing A549 cells with IC50 of 8.2 nM.
SM141 does not inhibit the other cysteine protease, papain-like protease (PLPro) and CatL.
SM141 reduced viral replication, viral loads in the lung, and enhanced survival in SARS-CoV2 infected K18-ACE2 transgenic mice.
