SOST inhibitor S6 is a small-molecule targeting Sclerostin (SOST), blocks STAT3 binding to SOST, inhibits breast cancer bone metastasis.
S6 treatment significantly inhibited the proliferation of SCP2 cells with an IC50 of 1.8 uM.
SOST inhibitor S6 significantly decreased STAT3 phosphorylation and TGF-β, KRAS, SMAD3 and CXCR4 expression in SCP2 cells.
S6 inhibited STAT3 phosphorylation and attenuated the downstream TGF-β/KRAS signaling pathways, which in turn inhibited breast cancer progression.
S6 treatment enhanced the differentiation of BMSCs, S6 (10 mg/kg) inhibited the destruction of bone cortex and reduced osteoclast activity in the tumor-bearing mice.
