Biological Activity
SPH7854 is a potent, selective gut-restricted RORγt antagonist with IC50 of 10 nM in RORγt binding assays, shows inhibitory activity against Th17 cell differentiation with IC50 of 34.4 nM.
SPH7854 displays excellent binding activity and selectivity for RORγ (IC50-13.7 nM) over RORα and RORβ (IC50>10000 nM).
SPH7854 effectively inhibited hPBMC Type 17 cell differentiation (IC50=147.4 nM).
SPH7854 significantly promoted the generation of Treg cells at a concentration of 3 μM and inhibited the differentiation of CD4+ inflammatory cells in rats.
SPH7854 (3mpk, bid, oral) effectively improved TNBS-induced colitis in TNBS-induced acute colitis mouse model.
