Biological Activity
SQ-030 is a potent, hepatocyte-specific cytosolic prostaglandin E synthase (cPGES) inhibitor with SPR KD of 0.315 uM.
SQ-030 significantly reduces lipid accumulation in PA + OA-treated AML12 cells, while showing no apparent cytotoxicity across a concentration range of 0.01-100 μM in AML12 cells.
SQ-030 markedly alleviates liver dysfunction (serum AST and ALT), hepatic steatosis, inflammation and fibrosis, as shown by histology and by the expression of inflammatory and fibrotic markers in MCD-fed mice.
Cytosolic prostaglandin E synthase (cPGES, encoded by PTGES3) is a key enzyme in PGE2 synthesis, regulates MASLD and the synthesis of PGE2.
