SR-3677 is a potent and selective ROCK-II inhibitor with biochemical IC50 of 3 nM, inhibits myosin light chain bisphosphorylation (ppMLC) with IC50 of 3.5 nM in cell-based assays.
SR-3677 weakly inhibits ROCK-I with IC50 of 56 nM, exhibits an off-target hit rate of 1.4% against 353 kinases.
SR-3677 is efficacious in both, increasing ex vivo aqueous humor outflow in porcine eyes and inhibiting myosin light chain phosphorylation.
