SY-5609 (SY5609) is a highly potent, selective, noncovalent and orally available inhibitor of CDK7 with Kd of 0.07 nM in SPR assays (binding to CDK7/Cyclin H).
SY-5609 demonstrates high selectivity over CDK2, CDK9, and CDK12 (IC50>1,500 nM).
SY-5609 potently inhibits HCC70 cell proliferation (EC50=1 nM), SY-5609 induces G2/M cell cycle arrest in and triggers apoptosis in cancer cell lines.
SY-5609 is capable of inhibiting CDK7 in vivo and demonstrates regression in murine xenograft models.
