Salvage-1 is a selective, positive allosteric modulator (PAM) of α4β2 nAChRs, demonstrates potent antiarrhythmic efficacy.
Salvage-1 suppresses re-entry through α4β2 nAChR-mediated enhancement of myocardial excitability and conductivity.
Salvage-1 selectively stabilizes the α4 subunit (CHRNA4), with minimal effect on the β2 subunit (CHRNB2), also shows weak inhibition of key cardiac ion channels, Cav1.2 and Nav1.5.
Salvage-1 selectively potentiates α4β2 nAChRs by interacting with residues Phe312 and Phe316 of α4 subunit, thereby stabilizing the receptor in its open-channel state.
Salvage-1 demonstrates marked antiarrhythmic efficacy in rodent models of fatal ventricular tachyarrhythmias (FVTs).
Salvage-1 demonstrates superior efficacy compared to lidocaine in both porcine and human models of FVTs.
