Sinomenine is a natural alkaloid with potential anti-fibrotic activity, inhibits the binding of Keap1 to Nrf2, promotes the nuclear accumulation of Nrf2, induces upregulated expressions of Nrf2-dependent antioxidative genes. Sinomenine regulates macrophage reprogramming via the PI3K-AKT-NF-kappaB pathway to ameliorate osteoarthritis. Sinomenine regulates the AKT/FOXO3/GLUL pathway to inhibit pulmonary fibroblast-to-myofibroblast transition via alpha7nAChR against rheumatoid arthritis associated interstitial lung disease. Sinomenine efficiently induces nuclear translocation of Nrf2, and increased in ARE activity in a concentration-dependent manner.
