TJ-M11 is a synthesized peptide inhibitor of YAP/TAZ transactivation domain (TAD), specifically blocks YAP transactivation (IC50=5.96 uM, NCI-H1299 cells) by disrupting its TAD interactions with the cofactors MED15 and TAF4.
TJ-M11 blocks the interactions of YAP with TAF4 and MED15 in cells through Co-immunoprecipitation.
TJ-M11 disrupts the transcription of YAP target genes, including the immunosuppressive genes CSF1, ITGB2, and IL33.
TJ-M11 exhibits biological activity against cancer cell growth, inhibits primary patient-derived lung adenocarcinoma (LUAD) cells (LUAD1, IC50=6.77 uM).
TJ-M11 treatment significantly suppressed the tumor size, reverses adaptive resistance of anti-PD-1 inhibitor in TDCL tumor-bearing C57BL/6 mice.
