Biological Activity
TRPV2 activator AV2-1 is a potent, selective TRPV2 activator, activates mTRPV2 (EC50=13.6 μM), hTRPV2 (EC50=51.8 μM) and rat TRPV2 (rTRPV2; EC50=36.3 μM).
AV2-1 shows no activity against thermoTRPV channels (TRPV1, TRPV3 and TRPV4), TRPM2 and TRPM8.
AV2-1 robustly activates TRPV2 at concentrations of 50 and 100 μM for HEKmTRPV2 and HEKhTRPV2, respectively, in fura-2-mediated single cell [Ca2+]i assays.
AV2-1 exerts low toxicity in HEKTRPV2 cells and in human peripheral blood-derived macrophages (hPBDM).
AV2-1 activates TRPV2 channels in human macrophages derived from peripheral blood.
AV2-1 induces mouse bone marrow-derived mast cells (mBMMCs) degranulation.
