UFMylation inhibitor CP-24 is a potent, non-covalent UFMylation inhibitor targeting DDRGK1, disrupts of E3 ligase complex components DDRGK1-UFL1 interaction, binds directly to the DDRGK1 PCI domain, effectively inhibits UFMylation.
CP-24 showed significant upregulation of ER stress-related genes compared to DMSO-treated U87 cells, including HSPA5, XBP1, ERN1, EIF2AK3, CANX, ATF4, and DDIT3.
CP-24 effectively reduces global UFMylation levels, decrease DDRGK1 protein levels, inhibits ER-phagy, and induce ER stress.
CP-24 impairs ER-phagy, induces ER stress, and exhibits robust anti-tumor effects in vitro and in multiple GBM mouse models.
