Biological Activity
WCDD115 is a potent indirect inhibitor of NRF2 expression with IC50 of 57 nM (inhibition of eYFP quantitation in H1299 NQO1-eYFP cells), inhibits hDHFR with IC50 of 144 nM, suppresses endogenous metrics of NRF2 signal transduction irrespective of KEAP1 expression.
WCDD115 suppresses endogenous NRF2 signaling and increases DHFR protein in NRF2-active KYSE70 and A549 cells.
WCDD115 suppresses the RNA level expression of NRF2, NQO1, GCLC, and SXRN1 with similar efficacy。
Inhibition of DHFR is required for WCDD115-mediated suppression of NRF2.
