Biological Activity
WEHI-P4 is a highly potent, specific SARS-CoV-2 papain-like protease (PLpro) inhibitor with IC50 of 19 nM and KD of 18 nM, cellular EC50 of 330 nM in in-cell FRET-based biosensor assays.
WEHI-P4 displayed an antiviral EC50 of 410 nM in Vero plaque live virus infection assays, does not require auxiliary Pgp inhibition for its antiviral efficacy.
WEHI-P4 inhibits SARS-CoV-2 and SARS-CoV, and surprisingly also shows weak activity (IC50=52 µM) against PL2pro of the αCoV NL63, which has only 20% sequence identity to SARS-CoV-2.
