Biological Activity

WJM664 is a small molecule inhibitor of P. falciparum malaria P-type ATPase 4 (PfATP4), inhibits P. falciparum 3D7 parasite growth with EC50 of 13 nM.
WJM664 is potent against both male and female gametes with EC50 of 0.18 and 0.25 uM in dual gamete formation assays.
WJM664 inhibited gamete development and blocked parasite transmission to mosquitoes but exhibited low efficacy in aPlasmodium berghei mouse model, which was attributed to ATP4 species differentiation and its moderate systemic exposure.
