Biological Activity
ZDLT-1 is potent HDAC inhibitor with high inhibitory activity for class I HDACs 1, 2, and 3 with IC50 of 2.2-3.4 nM, shows potent anti-proliferative activity against HCT116 cells with IC50 of 0.173 uM.
ZDLT-1 does not inhibit HDAC8 (IC50>600 nM).
ZDLT-1 also exhibits better inhibitory activity with an IC50 value of 6 nM for HDAC6 than SAHA (IC50 = 15 nM).
ZDLT-1 induces apoptosis and inhibits cell migration in HCT116 cells.
ZDLT-1 can suppresse CRC cell growth in vivo and in vitro by triggering HIF-1α/Caspase-3/Caspase-9 pathway in promoting apoptosis, and triggering NF-κB/GSDMD/GSDME pathway in inhibiting tumor inflammation.
