ABBV-318 is a small molecule voltage-gated sodium channel Nav1.7/ Nav1.8 blocker with IC50 of 3.8 and 2.8 uM for hNav1.7 and hNav1.8 in FRET assays, respectively.
ABBV-318 shows no activity (IC50 > 30 µM) at known cardiac liability channel Nav1.5, and lack of potency at Nav1.2, Nav1.4, and Nav1.6 (>30 µM for all 3 channels in whole-cell patch clamp EP assay).
ABBV-318 displays robust in vivo efficacy in both inflammatory and neuropathic rodent models of pain.
