ABL127 (ML174) is a potent, selective, covalent inhibitor of protein methylesterase 1 (PME-1) with IC50 of 10 nM; shows IC50s of 6.4 nM and 4.2 nM in HEK293T and MDA-MB-231 cells, respectively, decreases cell proliferation and invasive growth in vitro; shows selectivity at 1 uM against 30 serine hydrolase anti-targets; ML174 is highly active against PME-1 and can cause 85% reduction of demethylated PP2A, demonstrates greater potential for use in long time-course in situ studies.
